Capabilities - Gene to Structure

Sareum offers full Gene-to-Structure projects, allowing our customers to see exactly how their small-molecule ligands interact with their protein targets. Our knowledge-based high-throughput approach successfully delivers crystal structures within 1 to 6 months and is based on 4 key components:
Protein Expression
Using a combination of experience and computational analysis, multiple protein constructs are designed for expression. These are then expressed using our unique high-throughput baculoviral and / or E.coli expression systems. We typically make 96 versions of any candidate protein in each system in the first round of protein expression.
Protein Purification
The highly automated multi-column process allows rapid and thorough purification of multiple protein variants to carry through to the crystallisation platform.
Crystallisation
Our robotic crystallisation technology provides highly effective, flexible and productive screening and optimisation of crystallisation conditions. By carrying multiple versions of a protein through our process we determine the crystal form most suitable for the generation of high resolution protein-ligand complexes.
Structure Determination
After optimising cryopreservation, X-ray data is collected either on our in-house high resolution source or at one of a number of European synchrotron sources, allowing us to solve many crystal structures quickly and efficiently.
Our structural and molecular biologists have extensive expertise across a wide-range of protein families including kinases, nuclear hormone receptors, phosphodiesterases, proteases, transferases and polymerases. Most of these have been solved on an exclusive basis for our customers, but several are available on a non-exclusive basis via our Crystal Bank.
